In the presence of PAM in the target DNA, it is a RNA mediated Nuclease that specifically targets to cut double stranded DNA. The binding of LbCas12b crRNA complex with complementary single stranded DNA or double stranded DNA releases strong non-specific ssDNA transcleavage activity. Compared to Cas13a, Cas12b can maintain higher activity under loop mediated isothermal amplification (LAMP) reaction conditions.
